The leucine threshold is the minimum amount of leucine per meal required to reliably trigger a strong muscle protein synthesis (MPS) response. Practical value: typically around 2 to 3 g of leucine, usually achievable with roughly 25 to 40 g of high-quality protein per meal. For hardgainers this means: plan your protein pulses, don't just track daily totals.

Leucine threshold in 20 seconds

Leucine acts as the trigger amino acid and is a practical marker for "MPS on or off" per meal. What matters is leucine density per meal and distribution across the day, closely linked to protein quality, digestion and execution.

Timing guideline: leave roughly 3 to 5 hours between protein pulses so the next one can trigger MPS cleanly.

Where it sits in the system

  • Trigger amount: typically around 2 to 3 g of leucine per meal.
  • Mechanism: leucine is a signalling component in the MPS regulatory loop, without sufficient leucine concentration, the anabolic stimulus stays below the activation threshold.
  • Source matters: whey, dairy, eggs and meat are leucine-dense; plant-based sources work through combinations and slightly larger portions.

Leucine values are derived from typical protein content and the leucine share of that protein, roughly 8 to 11 percent for dairy and whey, roughly 7 to 8 percent for meat, egg and plants. These are reference values, not lab numbers for a specific product. The protein content is on the label, so you can do the same maths yourself.

Two things stand out. First, skyr and quark are excellent protein sources but sit far from the threshold per 100 g. A 250 g tub clears it, a spoonful in your porridge does not. Second, plant sources simply need more volume, and that is the real problem for a hard gainer: fullness arrives before the target does.

A plant-based setup becomes easier when you combine sources and keep your food hygiene consistent.

Practical setup in the Hardgainer system

  • 3 to 5 protein pulses: aim for 25 to 40 g of protein per meal as a base structure, don't bundle everything into one or two meals.
  • Couple with a lean surplus: the surplus sets the direction; leucine pulses determine the "quality" of the anabolic stimulus per meal.
  • Monitoring: 7-day average bodyweight, strength trend, digestion, sleep, don't overreact to single-day variation.

In practice: 14 days

The threshold is not a theory question, it is a distribution question. Two weeks are enough to see whether your meals clear it at all.

  • Day 0: Write down how many meals you eat on a normal day and how much protein sits in each. The label value is good enough.
  • Day 1 to 14: Flag every meal under 25 g of protein. That is roughly the line below which most sources do not clear the threshold.
  • Day 14, first question: How many meals per day were above it? At two out of four, half your meals run without a building stimulus.
  • Day 14, second question: Which meal is easiest to fix? Usually breakfast. That is where protein sits lowest and where it is cheapest to raise.

The usual finding is not too little protein per day, it is poor distribution. Eighty grams of protein with sixty of them at dinner does less than eighty spread evenly across four meals.

Where it goes wrong

  • Tracking only daily protein: MPS is triggered per meal. Distribution beats "everything in the evening", even at the same daily total.
  • Mini-snacks below the threshold: 10 to 15 g of protein often misses the leucine threshold. Consolidate portions or supplement with a leucine-dense source.
  • Training without nutritional context: without solid volume management and sufficient calories, protein pulses only get you so far.

Eggs and plant-based sources like lentils or tofu tend to require larger portions or combinations because their leucine content per 100 g of protein is somewhat lower. Three to five such protein pulses per day form a solid foundation.

Does the leucine threshold work with plant-based protein sources?

Yes, but with a little more planning. Plant-based sources generally have a lower leucine content per gram of protein. Through smart combinations and consistent food hygiene, the threshold is reliably achievable on a fully plant-based diet. Soya protein isolate offers the best leucine density among plant-based options.

How much leucine per meal in grams?

Roughly 2.5 g of leucine per meal is the figure used in practice. For most complete protein sources that lands at about 20 to 25 g of protein, since leucine makes up roughly 8 to 11 percent of the protein. With plant sources you need more, because the leucine share is lower and the protein content per 100 g is usually lower too.

How much leucine is in quark or skyr per 100 g?

Low-fat quark delivers about 1.15 g of leucine per 100 g, skyr about 1.05 g, derived from roughly 11 to 12 g of protein at a leucine share of about 9.5 percent. A 250 g tub therefore clears the threshold, a spoonful stirred into porridge does not.

The myth

If you want to build, crank the surplus and stop worrying about food quality. Calories are all that counts, the rest sorts itself out.

The reality

Calorie level and food quality are two separate dials. A moderate lean surplus of 150 to 250 kcal, four meals above the leucine threshold and a weekly weigh-in make building plannable. Clean bulk and dirty bulk do not differ in calorie amount, they differ in how much control you keep over it.

Four papers behind the claims on this page

The leucine threshold is not a switch, it is a dose-response observation. Four papers set the frame.

What the studies do not cover

None of these papers determined an exact leucine amount as a threshold. The widely cited 2.5 g per meal is derived from protein dose studies, not a directly measured cut-off. Whether a small meal below the threshold truly contributes nothing is therefore unanswered. Hard gainers were not studied as a separate group in any of them, and the table values are rounded reference numbers.

Practical takeaway: Four meals with at least 25 g of protein each from a complete source. Breakfast is almost always the meal with the biggest lever.
